Dual functionality of the amyloid protein TasA in Bacillus physiology and fitness on the phylloplane. Nat Commun. 2020 Apr 20;11(1):1859http://hdl.handle.net/10668/3676Bacteria can form biofilms that consist of multicellular communities embedded in an extracellular matrix (ECM). In Bacillus subtilis, the main protein component of the ECM is the functional amyloid TasA. Here, we study further the roles played by TasA in B. subtilis physiology and biofilm formation on plant leaves and in vitro. We show that ΔtasA cells exhibit a range of cytological symptoms indicative of excessive cellular stress leading to increased cell death. TasA associates to the detergent-resistant fraction of the cell membrane, and the distribution of the flotillin-like protein FloT is altered in ΔtasA cells. We propose that, in addition to a structural function during ECM assembly and interactions with plants, TasA contributes to the stabilization of membrane dynamics as cells enter stationary phase.enAtribución 4.0 Internacionalhttp://creativecommons.org/licenses/by/4.0/Bacillus subtilisBacterial proteinsBacteriaAmyloidogenic proteinsCell deathCell membraneProteínas bacterianasBacteriasProteínas amiloidogénicasMuerte celularMembrana celularMedical Subject Headings::Chemicals and Drugs::Amino Acids, Peptides, and Proteins::Proteins::Amyloid::Amyloidogenic ProteinsMedical Subject Headings::Organisms::Bacteria::Gram-Positive Bacteria::Bacillales::Bacillaceae::BacillusMedical Subject Headings::Organisms::Bacteria::Gram-Positive Bacteria::Bacillales::Bacillaceae::Bacillus::Bacillus subtilisMedical Subject Headings::Phenomena and Processes::Microbiological Phenomena::Bacterial Physiological Phenomena::Bacterial Processes::Bacterial AdhesionMedical Subject Headings::Chemicals and Drugs::Amino Acids, Peptides, and Proteins::Proteins::Bacterial ProteinsMedical Subject Headings::Phenomena and Processes::Microbiological Phenomena::BiofilmsMedical Subject Headings::Phenomena and Processes::Cell Physiological Phenomena::Cell Physiological Processes::Cell DeathMedical Subject Headings::Anatomy::Cells::Cellular Structures::Cell MembraneMedical Subject Headings::Organisms::Eukaryota::Plants::Viridiplantae::Streptophyta::Embryophyta::Angiosperms::CucurbitaceaeMedical Subject Headings::Disciplines and Occupations::Natural Science Disciplines::Biological Science Disciplines::Biology::EcologyMedical Subject Headings::Chemicals and Drugs::Lipids::LipopeptidesMedical Subject Headings::Phenomena and Processes::Genetic Phenomena::Genetic Variation::MutationMedical Subject Headings::Phenomena and Processes::Genetic Phenomena::Genetic Processes::Gene Expression::Transcription, Genetic::TranscriptomeDual functionality of the amyloid protein TasA in Bacillus physiology and fitness on the phylloplaneresearch article32313019Acceso abierto10.1038/s41467-020-15758-z2041-1723PMC7171179
