Pyrosequencing analysis reveals changes in intestinal microbiota of healthy adults who received a daily dose of immunomodulatory probiotic strains. Nutrients. 2015 ; 7(6):3999-4015http://hdl.handle.net/10668/2346Journal Article; Randomized Controlled Trial; Research Support, Non-U.S. Gov't;The colon microbiota plays a crucial role in human gastrointestinal health. Current attempts to manipulate the colon microbiota composition are aimed at finding remedies for various diseases. We have recently described the immunomodulatory effects of three probiotic strains (Lactobacillus rhamnosus CNCM I-4036, Lactobacillus paracasei CNCM I-4034, and Bifidobacterium breve CNCM I-4035). The goal of the present study was to analyze the compositions of the fecal microbiota of healthy adults who received one of these strains using high-throughput 16S ribosomal RNA gene sequencing. Bacteroides was the most abundant genus in the groups that received L. rhamnosus CNCM I-4036 or L. paracasei CNCM I-4034. The Shannon indices were significantly increased in these two groups. Our results also revealed a significant increase in the Lactobacillus genus after the intervention with L. rhamnosus CNCM I-4036. The initially different colon microbiota became homogeneous in the subjects who received L. rhamnosus CNCM I-4036. While some orders that were initially present disappeared after the administration of L. rhamnosus CNCM I-4036, other orders, such as Sphingobacteriales, Nitrospirales, Desulfobacterales, Thiotrichales, and Synergistetes, were detected after the intervention. In summary, our results show that the intake of these three bacterial strains induced changes in the colon microbiota.enGutHealthyAdultsHigh-throughput nucleotide sequencingMicrobiotaProbioticsADN bacterianoMicrobioma gastrointestinalVoluntarios sanosSecuenciación de nucleótidos de alto rendimientoFilogeniaProbióticosARN ribosómico 16SAnálisis de secuencia de ADNMedical Subject Headings::Organisms::Bacteria::Gram-Positive Bacteria::Actinobacteria::BifidobacteriumMedical Subject Headings::Chemicals and Drugs::Nucleic Acids, Nucleotides, and Nucleosides::Nucleic Acids::DNA::DNA, BacterialMedical Subject Headings::Anatomy::Fluids and Secretions::FecesMedical Subject Headings::Check Tags::FemaleMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Investigative Techniques::Genetic Techniques::Sequence Analysis::High-Throughput Nucleotide SequencingMedical Subject Headings::Organisms::Eukaryota::Animals::Chordata::Vertebrates::Mammals::Primates::Haplorhini::Catarrhini::Hominidae::HumansMedical Subject Headings::Anatomy::Digestive System::Gastrointestinal Tract::IntestinesMedical Subject Headings::Organisms::Bacteria::Gram-Positive Bacteria::Gram-Positive Rods::Gram-Positive Asporogenous Rods::Gram-Positive Asporogenous Rods, Regular::Lactobacillaceae::LactobacillusMedical Subject Headings::Organisms::Bacteria::Gram-Positive Bacteria::Gram-Positive Rods::Gram-Positive Asporogenous Rods::Gram-Positive Asporogenous Rods, Regular::Lactobacillaceae::Lactobacillus::Lactobacillus rhamnosusMedical Subject Headings::Check Tags::MaleMedical Subject Headings::Phenomena and Processes::Genetic Phenomena::PhylogenyMedical Subject Headings::Technology, Industry, Agriculture::Food and Beverages::Food::Dietary Supplements::ProbioticsMedical Subject Headings::Chemicals and Drugs::Nucleic Acids, Nucleotides, and Nucleosides::Nucleic Acids::RNA::RNA, Ribosomal::RNA, Ribosomal, 16SMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Investigative Techniques::Genetic Techniques::Sequence Analysis::Sequence Analysis, DNAMedical Subject Headings::Named Groups::Persons::Age Groups::Adult::Young AdultMedical Subject Headings::Named Groups::Persons::Age Groups::AdultMedical Subject Headings::Phenomena and Processes::Microbiological Phenomena::Microbiota::Gastrointestinal MicrobiomeMedical Subject Headings::Named Groups::Persons::Research Subjects::Healthy VolunteersPyrosequencing analysis reveals changes in intestinal microbiota of healthy adults who received a daily dose of immunomodulatory probiotic strains.research article26016655open access10.3390/nu70639992072-6643PMC4488769
