Real-time molecular epidemiology of tuberculosis by direct genotyping of smear-positive clinical specimens. J. Clin. Microbiol.. 2012 May; 50(5):1755-70095-1137http://hdl.handle.net/10668/2018Evaluation Studies; Journal Article; Research Support, Non-U.S. Gov't;We applied MIRU-VNTR (mycobacterial interspersed repetitive-unit-variable-number tandem-repeat typing) to directly analyze the bacilli present in 61 stain-positive specimens from tuberculosis patients. A complete MIRU type (24 loci) was obtained for all but one (no amplification in one locus) of the specimens (98.4%), and the allelic values fully correlated with those obtained from the corresponding cultures. Our study is the first to demonstrate that real-time genotyping of Mycobacterium tuberculosis can be achieved, fully transforming the way in which molecular epidemiology techniques can be integrated into control programs.enADN bacterianoTipificación molecularEsputoTuberculosis pulmonarMedical Subject Headings::Chemicals and Drugs::Nucleic Acids, Nucleotides, and Nucleosides::Nucleic Acids::DNA::DNA, BacterialMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Investigative Techniques::Genetic Techniques::Molecular TypingMedical Subject Headings::Organisms::Bacteria::Gram-Positive Bacteria::Gram-Positive Rods::Gram-Positive Asporogenous Rods::Gram-Positive Asporogenous Rods, Regular::Mycobacteriaceae::Mycobacterium::Mycobacterium tuberculosisMedical Subject Headings::Diseases::Respiratory Tract Diseases::Respiratory Tract Infections::Tuberculosis, PulmonaryMedical Subject Headings::Phenomena and Processes::Chemical Phenomena::Biochemical Phenomena::Molecular Structure::Base Sequence::Repetitive Sequences, Nucleic Acid::Tandem Repeat Sequences::Minisatellite RepeatsMedical Subject Headings::Organisms::Eukaryota::Animals::Chordata::Vertebrates::Mammals::Primates::Haplorhini::Catarrhini::Hominidae::HumansMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Investigative Techniques::Epidemiologic Methods::Molecular EpidemiologyMedical Subject Headings::Anatomy::Fluids and Secretions::Bodily Secretions::SputumReal-time molecular epidemiology of tuberculosis by direct genotyping of smear-positive clinical specimens.research article22378907open access10.1128/JCM.00132-121098-660X
