Doxycycline-Doped Polymeric Membranes Induced Growth, Differentiation and Expression of Antigenic Phenotype Markers of Osteoblasts. Polymers. 2021 Mar 28;13(7):1063http://hdl.handle.net/10668/4443Polymeric membranes are employed in guided bone regeneration (GBR) as physical barriers to facilitate bone in-growth. A bioactive and biomimetic membrane with the ability to participate in the healing and regeneration of the bone is necessary. The aim of the present study was to analyze how novel silicon dioxide composite membranes functionalized with zinc or doxycycline can modulate the osteoblasts' proliferation, differentiation, and expression of selected antigenic markers related to immunomodulation. Nanostructured acrylate-based membranes were developed, blended with silica, and functionalized with zinc or doxycycline. They were subjected to MG63 osteoblast-like cells culturing. Proliferation was assessed by MTT-assay, differentiation by evaluating the alkaline phosphatase activity by a spectrophotometric method and antigenic phenotype was assessed by flow cytometry for selected markers. Mean comparisons were conducted by one-way ANOVA and Tukey tests (p < 0.05). The blending of silica nanoparticles in the tested non-resorbable polymeric scaffold improved the proliferation and differentiation of osteoblasts, but doxycycline doped scaffolds attained the best results. Osteoblasts cultured on doxycycline functionalized membranes presented higher expression of CD54, CD80, CD86, and HLA-DR, indicating a beneficial immunomodulation activity. Doxycycline doped membranes may be a potential candidate for use in GBR procedures in several challenging pathologies, including periodontal disease.enAtribución 4.0 Internacionalhttp://creativecommons.org/licenses/by/4.0/CD markersDoxycyclineAntigenic phenotypeOsteoblastsMembraneAlkaline phosphataseFlow cytometryBone regenerationCell proliferationPeriodontal diseaseImmunomodulationAntígenos CDDoxiciclinaOsteoblastosFosfatasa alcalinaCitometría de flujoRegeneración óseaProliferación celularEnfermedades periodontalesInmunomodulaciónMembranasMedical Subject Headings::Chemicals and Drugs::Enzymes and Coenzymes::Enzymes::Hydrolases::Esterases::Phosphoric Monoester Hydrolases::Alkaline PhosphataseMedical Subject Headings::Chemicals and Drugs::Organic Chemicals::Hydrocarbons::Hydrocarbons, Cyclic::Hydrocarbons, Aromatic::Polycyclic Hydrocarbons, Aromatic::Naphthacenes::Tetracyclines::DoxycyclineMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Diagnosis::Diagnostic Techniques and Procedures::Clinical Laboratory Techniques::Cytological Techniques::Cell Separation::Flow CytometryMedical Subject Headings::Disciplines and Occupations::Natural Science Disciplines::Biological Science Disciplines::Biotechnology::BiomimeticsMedical Subject Headings::Phenomena and Processes::Musculoskeletal and Neural Physiological Phenomena::Musculoskeletal Physiological Phenomena::Musculoskeletal Physiological Processes::Bone Remodeling::Bone RegenerationMedical Subject Headings::Anatomy::Cells::Connective Tissue Cells::OsteoblastsMedical Subject Headings::Phenomena and Processes::Cell Physiological Phenomena::Cell Physiological Processes::Cell Growth Processes::Cell ProliferationMedical Subject Headings::Chemicals and Drugs::Inorganic Chemicals::Minerals::Silicon DioxideMedical Subject Headings::Chemicals and Drugs::Amino Acids, Peptides, and Proteins::Proteins::Glycoproteins::Membrane Glycoproteins::Histocompatibility Antigens Class II::HLA-D Antigens::HLA-DR AntigensMedical Subject Headings::Diseases::Stomatognathic Diseases::Mouth Diseases::Periodontal DiseasesMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Therapeutics::Biological Therapy::ImmunomodulationMedical Subject Headings::Technology and Food and Beverages::Technology, Industry, and Agriculture::Manufactured Materials::Nanostructures::NanoparticlesMedical Subject Headings::Phenomena and Processes::Genetic Phenomena::PhenotypeMedical Subject Headings::Chemicals and Drugs::Organic Chemicals::Carboxylic Acids::Acids, Acyclic::AcrylatesMedical Subject Headings::Analytical, Diagnostic and Therapeutic Techniques and Equipment::Investigative Techniques::Epidemiologic Methods::Statistics as Topic::Analysis of VarianceMedical Subject Headings::Chemicals and Drugs::Inorganic Chemicals::Metals::Metals, Heavy::ZincMedical Subject Headings::Technology and Food and Beverages::Technology, Industry, and Agriculture::Manufactured Materials::Biomimetic Materials::Membranes, ArtificialDoxycycline-Doped Polymeric Membranes Induced Growth, Differentiation and Expression of Antigenic Phenotype Markers of Osteoblastsresearch article33800569Acceso abierto10.3390/polym130710632073-4360PMC8037272
